Target Information

FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead family. It functions as both coactivator and corepressor of nuclear receptor activity through the LXXLL motif in its C-terminal region. Association of FOXO1 with PAX3 has been linked to alveolar rhabdomyosarcoma. FOXO1 plays roles in autophagy, myogenic differentiation, and tumor suppression. It is a key target of insulin signaling, regulating metabolic homeostasis and oxidative stress response. FOXO1 binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE). It regulates redox balance, osteoblast numbers, bone mass, and endocrine function of the skeleton in glucose metabolism. In hepatocytes, it promotes gluconeogenesis by activating genes such as IGFBP1, G6PC, and PPCK1 in cooperation with PPARGC1A. FOXO1 also acts downstream of CDK1, PKB/AKT1, and SKT4/MST1 in regulating cell death.
