Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor within the TNFR (tumor necrosis factor receptor) superfamily. It also exists in a soluble form (~26 kDa).
Activation of FAS leads to aggregation of its intracellular FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and subsequent caspase activation.
This cascade triggers apoptosis and can also activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK pathways, contributing to proliferative signaling in fibroblasts and T cells.
At least eight alternatively spliced transcript variants encoding seven distinct isoforms have been described; isoforms lacking the transmembrane domain may negatively regulate apoptosis.
The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
