Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. It also exists in a soluble form (~26 kDa). Stimulation of FAS triggers aggregation of FADD, formation of the death-inducing signaling complex (DISC), and activation of caspases, initiating apoptosis. FAS can activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK pathways, and is involved in proliferative signaling in fibroblasts and T cells. Multiple splice variants exist, with some isoforms regulating apoptosis negatively. The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a role in antiviral immune responses.
