Product Specific Information

This 19-3 monoclonal antibody recognizes human and mouse terminal deoxynucleotidyl transferase (TdT), a 60 kDa polymerase responsible for the template-independent addition of N-nucleotides at gene segment junctions of developing lymphocytes.
Expression is found in the nucleus of immature lymphocytes but not in mature lymphocytes or non-lymphoid cells. TdT plays a critical role during TCR and Ig gene rearrangement. Expression in the mouse thymus correlates with T cell selection and is decreased after positive selection. TdT is also present in lymphoma or lymphoblastic leukemia cells.

Applications Reported:
For intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5521-00) for intracellular staining of mouse thymocytes. Recommended usage ≤ 1.0 µg/test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

Terminal Deoxynucleotidyl Transferase (TdT) is a DNA polymerase located in the nucleus that catalyzes the polymerization of deoxynucleotides at the 3′ hydroxyl ends of oligo- or polydeoxynucleotide initiators, functioning without a template.
TdT serves as a specific marker for diagnosing and classifying acute lymphoblastic lymphomas/leukemias, particularly useful in differentiating lymphoblastic lymphoma from Burkitt’s lymphoma.
