Target Information
ERN1 serine/threonine kinase (also known as endoplasmic reticulum to nucleus signaling 1, IRE1α) is a human homologue of the yeast Ire1 gene product.
It possesses intrinsic kinase and endoribonuclease activity and plays a key role in ER-based stress signaling.
ERN1 regulates IRE1 proteolysis in mammalian cells and is critical for extraembryonic cell function essential for fetal viability.
IRE1 acts as a sensor of unfolded proteins in the ER, initiating signaling to the nucleus by splicing XBP1 mRNA, converting it into a potent unfolded-protein response transcriptional activator.
Null mutants are viable, but IRE1 is essential for survival under ER stress conditions. Dominant-negative forms of IRE1p do not affect normal cells but induce apoptosis under unfolded protein stress.
