Product Specific Information
PA1-032 detects phosphorylated nNOS (Ser1417) from rat and human samples.
It has been successfully used in Western blot and immunofluorescence procedures.
By Western blot, this antibody detects an approximately 140 kDa protein representing recombinant rat phospho-nNOS (Ser1417).
The immunogen is a synthetic phosphopeptide corresponding to residues C N(1411) R L R S E (pS) I A F I E E S K(1425) of human nNOS.
The peptide sequence is 100% conserved in rat, frog, and mouse proteins.
This peptide (Cat. # PEP-188) is available for use in neutralization and control experiments.
Target Information
NOS (Nitric Oxide Synthase) activity requires homodimerization and three cosubstrates (L-arginine, NADPH, and O₂), as well as five cofactors or prosthetic groups (FAD, FMN, calmodulin, tetrahydrobiopterin, and heme).
Three distinct NOS isoforms have been identified, encoded by separate genes:
- NOS1 (nNOS or ncNOS): Neuronal form, Ca²⁺/CaM-dependent
- NOS2 (iNOS): Inducible form
- NOS3 (eNOS or ecNOS): Endothelial form, Ca²⁺/CaM-dependent
