Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Interferon gamma (IFNγ, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines.
It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
Biological roles include:
- Antiviral and tumor antiproliferative activity
- Induction of class I and II MHC expression
- Macrophage activation
- Enhanced immunoglobulin secretion by B lymphocytes
Human IFNγ is species-specific and does not cross-react with mouse IFNγ.
It acts via IFNγ receptor I and II, activating the JAK-STAT pathway.
Expression is upregulated by IL-2, FGF basic, and EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFNγ gene are associated with aplastic anemia.
