Product Specific Information
- For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Interferon gamma (IFNγ), also known as Type II interferon, is a macrophage activation factor produced primarily by T-lymphocytes and natural killer cells in response to antigens and cytokines.
It is a dimeric glycoprotein (approx. 45 kDa) composed of two 146 amino acid subunits.
On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
The biological activity of IFNγ is highly species-specific, with no cross-reactivity between human and mouse IFNγ.
Functions:
- Antiviral and tumor antiproliferative activity
- Induction of class I and II MHC expression
- Macrophage activation
- Enhancement of immunoglobulin secretion by B lymphocytes
- Regulation of cytokine expression and synergistic action with other cytokines
Activation occurs via binding to IFNγ receptor I and II, initiating the JAK-STAT signaling pathway.
Human IFNγ shares ~40% sequence homology with mouse IFNγ.
Expression is upregulated by IL-2, FGF basic, and EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFNγ gene are associated with aplastic anemia.
For Research Use Only. Not for use in diagnostic procedures. Not for resale without express authorization.
