Product Specific Information
- For Immunohistochemistry (PFA-fixed):
Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Interferon gamma (IFNγ, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines.
It is a dimeric glycoprotein (~45 kDa) composed of two 146-amino-acid subunits, functioning as a homodimer.
On SDS-PAGE, IFNγ appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
Human IFNγ shows species specificity and does not cross-react with mouse IFNγ.
Functions include:
- Antiviral and tumor antiproliferative activity
- Induction of MHC class I and II
- Macrophage activation
- Enhancement of immunoglobulin secretion by B cells
- Cytokine regulation and synergistic effects with other cytokines
Activation occurs via IFNγ receptor I/II and the JAK-STAT pathway.
Human IFNγ shares ~40% sequence homology with mouse IFNγ.
Expression is upregulated by IL-2, FGF-basic, and EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFNγ gene are associated with aplastic anemia.
