Target Information

Metadherin (MTDH, LYRIC) is a metastasis adhesion protein associated with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is not a structural component but is recruited during tight junction maturation.
Metadherin is overexpressed in breast cancer tissue and xenografts, with low expression in normal breast tissue. It binds to lung vasculature—a common site of breast cancer metastasis—via a C-terminal extracellular domain segment. Blocking this domain or inhibiting metadherin with siRNA has been reported to reduce breast cancer metastasis.
