Efficient Generation of Edited hiPS Clonal Populations

Establishing clonal populations from edited hiPS cells grown and passaged as colonies is inefficient and time-consuming, often resulting in cell death or premature differentiation.
The Cellartis iPSC Single-Cell Cloning DEF-CS Culture Media Kit provides a complete system for efficient expansion and scale-up manufacturing of hiPS cells in a feeder-free and defined environment.

This kit includes all necessary reagents for:

Seeding single hiPS cells into 96-well plates
Expanding clones into 48-well plates
Continued culture using the Cellartis DEF-CS 500 Culture System

To generate edited clonal hiPS cell lines from start to finish, combine this kit with an electroporation-based or gesicle-based Cas9-sgRNA RNP complex delivery system.

Overview

Superior single-cell survival (~50%) when seeded in 96-well plates
Maintenance of pluripotency (>90%) for Oct-4, TRA-1-60, and SSEA-4 markers
Stable karyotype maintained throughout cloning and expansion
Compatible with CRISPR/Cas9 gene-editing via electroporation or gesicle-based delivery
