• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

BCL-2 is a key regulator of apoptosis that can inhibit or promote cell death. Members of the BCL-2 family form dimers to modulate apoptosis.

• Bag1 forms heterodimers with BCL-2, enhancing its anti-apoptotic effect.
• Bax and Bak are critical for cytochrome c release from mitochondria and initiate apoptosis.
• Constitutive expression of BCL-2, often due to translocation to the Ig heavy chain locus, is associated with follicular lymphoma.
• In follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, while normal or hyperplastic centers do not.
• Two transcript variants exist, differing in their C-terminal ends.
• Overexpression of BCL-2 has been linked to cancers such as B-cell lymphoma and prostate cancer.
