Staining of formalin-fixed tissues requires digesting tissue sections with Pepsin for 10 minutes at 37°C, followed by incubation with primary antibody for 10 minutes at room temperature.
A suggested positive control is skin or squamous cell carcinoma.

Target Information

Cytokeratins (CK) are intermediate filaments of epithelial cells, found in both keratinizing tissues (e.g., skin) and non-keratinizing cells (e.g., mesothelial cells).
At least 20 different CKs can be identified, which are classified into two groups:

• Type I (acidic polypeptides)
• Type II (basic polypeptides)

Keratin 14, a type I low molecular weight keratin, pairs with keratin 5 and serves as a marker distinguishing stratified epithelial cells from simple epithelial cells.
It is useful in identifying squamous cell carcinomas and is considered a prognostic marker in breast carcinomas.
Both acidic and basic keratin types are expressed in all epithelial cells.
