Product Specific Information

The polyclonal antibody reacts with mouse, rat, and human NFkB p65; it was raised against a peptide mapping to the carboxy terminus of human NFkB p65. Members of the Rel/NFkB family of transcription factors regulate cellular processes such as growth, development, and inflammation. They contain a Rel homology region (RHR) mediating protein dimerization. Complexes of p50 (NF-kB1) or p52 (NF-kB2) associate with Rel family members (p65, c-Rel, RelB) to regulate transcription of different genes.

In resting cells, NFkB is retained in the cytoplasm by IkB proteins. Upon activation by cytokines, LPS, or oxidative stress, IkB is degraded, allowing NFkB to translocate to the nucleus and activate proinflammatory genes. NFkB activation is implicated in various pathological conditions, including cardiac ischemia-reperfusion injury and insulin-dependent diabetes mellitus.

Target Information

Nuclear factor kB p65 (NF-kB p65), encoded by the RELA gene on human chromosome 11, is also known as RelA. It forms heterodimers with NF-kB subunits p50 or p52 and plays a key role in the transcriptional regulation of immune and inflammatory genes. Activation occurs via TNFα-induced IKK phosphorylation, leading to IκB degradation and nuclear translocation of NFkB. It is critical for immunoglobulin κ gene transcription in mature B lymphoid cells.
