Target Information
Transglutaminase (TGase)는 Ca²⁺ 결합을 필요로 하며, 단백질 간 ε-(γ-glutamyl) lysine 이소펩타이드 결합을 형성하여 단백질 교차결합을 촉매합니다.
포유류에서는 8종의 TGase 동종효소가 확인되었으며, Tissue transglutaminase (tTGase, TGase 2)는 N-말단 β-sandwich, 촉매 코어, 그리고 2개의 C-말단 β-barrel 도메인으로 구성되어 있습니다.
tTGase는 세포사멸, 증식, 분화, 수용체 매개 엔도사이토시스 등 다양한 세포 기능에 관여할 수 있습니다.
알츠하이머병 뇌에서는 tTGase 활성 증가로 인해 Ab peptide, β-amyloid precursor protein, tau protein 등의 중합이 일어나며, 이는 신경섬유 엉킴(neurofibrillary tangles) 형성과 관련이 있습니다.
