Target Information
STAT3는 STAT (Signal Transducers and Activators of Transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동한 후 전사 인자로 작용합니다. 이 단백질은 사이토카인 및 성장 인자에 반응하여 전사 활성화를 조절하며, CREB-binding protein 등의 공동활성화 인자가 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL-6 등에 의해 활성화될 수 있습니다. Tyr705 잔기의 인산화는 STAT3 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다. 인간에서 STAT3 유전자는 염색체 17q에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다.
STAT3는 세포 성장, 분화, 세포사멸 조절 등 다양한 세포 과정에서 중요한 역할을 합니다. Rac1 GTPase는 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제 인자로 알려져 있습니다. 세 가지 전사체 변이형이 보고되었으며, STAT3 유전자 결실 마우스는 초기 배아 단계에서 치명적입니다.
