Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 Alzheimer’s disease 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다. Abeta는 40–43개의 아미노산으로 구성되며, beta-amyloid precursor protein (beta APP)의 절단을 통해 생성됩니다.
첫 번째 절단은 beta-secretase (BACE)에 의해 수행되며, 이는 prohormone convertase인 furin에 의해 활성화됩니다. 이어서 gamma-secretase 복합체(주요 구성 요소로 presenilin-1, presenilin-2, nicastrin 포함)에 의해 추가 절단이 이루어져 amyloid beta peptide가 생성됩니다.
Abeta는 신경세포 외부의 섬유성 단백질로서 amyloid cores, neuritic plaques, neurofibrillary tangles 등에 존재하며, Alzheimer’s disease뿐 아니라 Lewy body dementia, Down’s syndrome, cerebral amyloid angiopathy 등에서도 발견됩니다.
