This antibody detects endogenous levels of total IKK alpha/beta.
Target Information
IKK alpha/beta is encoded by CHUK (conserved helix-loop-helix ubiquitous kinase) and IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) genes located on chromosomes 10 and 8 respectively in humans.
Upon stimulation by agents that activate the NF-κB pathway such as TNF-alpha, the two subunits of the IKK complex (IKK alpha and IKK beta) together with the regulator NEMO (IKK gamma) phosphorylate IκB proteins. This leads to ubiquitin-mediated degradation of IκB and phosphorylation of NF-κB, resulting in its translocation to the nucleus.
Phosphorylation at serine residues 176 and 180 in the activation loop induces a conformational change required for kinase activation.
