Target Information

Ninein is a component of the subdistal appendages of the mother centriole, localized specifically in the pericentriolar matrix of the centrosome. It may be involved in microtubule minus-end capping, centriole positioning, and protein anchoring. Studies indicate that microtubule nucleation and anchoring at the centrosome are independent processes linked by ninein function. Multiple human and mouse isoforms have been reported, with evidence suggesting that combined C-terminal and N-terminal domains can localize ninein to the mother centriole even in the absence of a coiled-coil region. Microinjection of anti-ninein antibodies into HeLa cells disrupts centrosomal reformation, leading to dispersal of centrosomal material. Such antibodies can also interfere with MTOC function and are found in sera from patients with autoimmune diseases targeting centrosomal proteins.
