Target Information

Tight junctions form an important barrier for paracellular transport in epithelial cells. Tricellulin is the first protein identified to specifically concentrate in tricellular tight junctions, where three adjacent cells meet. It possesses four membrane-spanning domains similar to claudins and is highly expressed in epithelial tissues such as small intestine, kidney, and lung.
Functional studies using siRNA show that suppression of tricellulin expression compromises epithelial barrier integrity and tight junction formation. Mutations in tricellulin that remove conserved regions in its cytosolic domain, which interacts with scaffolding protein ZO-1, highlight its critical role in tight junction structure and function.
