Product Specific Information

Description:
The 2D10.4 monoclonal antibody reacts with human CD80 (B7-1), a 60 kDa member of the Ig superfamily. CD80 is expressed by activated B cells, macrophages, dendritic cells, and activated T cells. It binds with high affinity to CD28 and CD152 (CTLA-4) on T cells, playing a critical role in T-B cell communication and activation.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested on Daudi cell line for flow cytometry. Recommended usage: 5 µL (1.0 µg) per test (100 µL volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass.

Staining Recommendations:
For panels with multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

  • IC Fixation Buffer ([Product # 00-8222-49])
  • 1-step Fix/Lyse Solution ([Product # 00-5333-57])
    Clone-specific performance should be validated empirically.

Target Information

CD80 (B7-1) and CD86 (B7-2) are ligands of T cell costimulatory molecule CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on antigen-presenting cells and are essential for T cell activation.
CD80 expression is rapidly induced in activated B cells, EBV-transformed B cell lines, Burkitt’s lymphoma cells, follicular B lymphoma, T cells, and monocytes.
CD80 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
