Product Specific Information

The 9F10 monoclonal antibody reacts with human CD49d, the 150 kDa integrin alpha4 subunit. CD49d forms a complex with integrin beta1 (CD29), known as VLA-4, which serves as a receptor for fibronectin and VCAM-1 (CD106). This complex is expressed on thymocytes, peripheral lymphocytes, monocytes, and eosinophils. CD49d also associates with integrin beta7 and binds to Mucosal Addressin Cell-Adhesion Molecule-1 (MadCAM-1).

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated with normal human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light during storage and use.

Target Information

Integrin alpha 4 (CD49d, ITGA4) is a member of the integrin alpha chain family. Integrins are heterodimeric membrane proteins composed of alpha and beta subunits. Alpha4 associates with either beta1 or beta7 chains. Ligands include MadCAM, VCAM, and fibronectin. MadCAM is the principal ligand for integrin alpha4beta7. HIV-1 envelope can mimic MadCAM by binding and signaling through integrin alpha4beta7, the gut mucosal homing receptor for peripheral T cells.
