Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen (also known as BCM1, Blast-1, and OX-45). CD48, a 45 kDa GPI-linked glycoprotein of the SLAM and Ig superfamily, is expressed on most hematopoietic cells. It plays a critical role in adhesion and T cell activation.

Hematopoietic stem cells (HSC) are characterized as CD150(+)CD244(-)CD48(-), while multipotent progenitors (MPP) are CD244(+)CD150(-)CD48(-), and restricted progenitors are CD48(+)CD244(+)CD150(-). HM48-1 modulates CD48 functions by blocking CD48/CD2 and CD48/CD244 interactions, inhibiting mitogen-induced proliferation, providing costimulatory signals for T cells, and prolonging cardiac allograft survival.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended detection filter: 710/50 bandpass.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD48 (Blast-1) is a GPI-anchored protein of the Ig superfamily, expressed broadly on hematopoietic cells. It serves as a high-affinity ligand for 2B4 and a low-affinity ligand for CD2. The 2B4-CD48 interaction regulates NK and NK-T cell proliferation and eosinophil activation. CD48 expression increases in several infectious diseases.

It is strongly expressed on lymphocytes and monocytes, weakly on granulocytes, and absent on platelets, fibroblasts, epithelial, and endothelial cells. CD48 is a marker for detecting GPI-anchor defects in paroxysmal nocturnal hemoglobinuria (PNH) and may be involved in allergic airway disease pathology.
