Description:
The OX8 monoclonal antibody reacts with rat CD8 alpha, a member of the immunoglobulin superfamily that heterodimerizes with CD8 beta to form the CD8 antigen. CD8 is expressed on CD4+CD8+ thymocytes, MHC class I-restricted peripheral T cells, and some macrophages. It binds to MHC class I molecules on antigen-presenting cells and participates in T-cell receptor signal transduction via Lck kinase.

Applications Reported:
This antibody is suitable for flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.25 µg per test (in 100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, functioning as a co-receptor with TCR for MHC class I molecules. It is mainly expressed as a disulfide-linked heterodimer (CD8α/β) on T cells and as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
Upon MHC-I/peptide binding, CD8 recruits Lck kinase, triggering lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
