MA1-752 detects presenilin 1 protein (PS1) from mouse, rat, human, and nonhuman primate samples.
No cross-reactivity is observed with presenilin 2.

MA1-752 has been successfully used in:

• Immunofluorescence
• Immunocytochemistry
• Western blot
• Immunoprecipitation
• ELISA

By Western blot, this antibody detects an ~18 kDa PS1 C-terminal fragment and the ~46 kDa full-length PS1 from transfected SH-SY5Y cells.
In 4% paraformaldehyde-fixed N2a cells, MA1-752 showed endogenous PS1 staining in the Golgi and cellular vesicles.

The immunogen is a synthetic peptide corresponding to residues C S(313) K Y N A E S T E R E S Q D T V A E N D D G(334) of human PS1.

Target Information

Presenilin 1 (PS1) was initially identified as a marker of susceptibility to early-onset Alzheimer’s disease.
Along with PEN2, nicastrin, and APH-1, PS1 forms the γ-secretase protein complex—a membrane-bound aspartyl protease that cleaves certain proteins within the lipid bilayer.

This cleavage plays a crucial role in:

• Cell-surface receptor signaling
• Generation of neurotoxic amyloid peptides linked to Alzheimer’s disease

γ-secretase cleaves the amyloid precursor protein (APP) within its intramembrane region, producing either the non-toxic p3 peptide or the toxic Aβ peptide.
Accumulation of Aβ peptide is considered a precursor to Alzheimer’s disease.
Multiple isoforms of presenilin 1 are known to exist.
