Product Specific Information

The A2MR-a2 monoclonal antibody reacts with human CD91, also known as LRP-1 (alpha-2 macroglobulin receptor).
CD91 is a 600 kDa type I transmembrane protein composed of alpha and beta chains. The alpha chain binds various ligands, while the beta chain contains tyrosine phosphorylation motifs that interact with adaptor and signaling proteins. Ligand binding mediates endocytosis and lysosomal trafficking.

CD91 is expressed in monocytic lineage cells, dendritic cells, hepatocytes, fibroblasts, adipocytes, neurons, and syncytiotrophoblasts. It plays roles in:

• Antigen cross-presentation via MHC class I molecules
• Cell migration and activation of signaling cascades

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.125 µg)/test.

A test is defined as the amount of antibody staining a sample in 100 µL final volume. Cell number may vary from 10⁵ to 10⁸ cells/test.

Super Bright 600 is a tandem dye excited at 405 nm and emitting at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and samples from light.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer ([Product # 00-8222-49])
• 1-step Fix/Lyse Solution ([Product # 00-5333-57])

Target Information

The CD91 gene encodes an endocytic receptor involved in:

• Intracellular signaling and lipid homeostasis
• Clearance of apoptotic cells
• A2M-mediated clearance of amyloid precursor protein and beta-amyloid (Alzheimer’s disease relevance)

Expression decreases with age and is reduced in Alzheimer’s patient brain tissue.
