Product Specific Information

The A2F10 monoclonal antibody reacts with mouse CD135 (Flk2/Flt3), a 135–150 kDa tyrosine kinase receptor expressed by primitive progenitor cells in fetal liver and adult bone marrow. Two-color staining with A2F10 and markers such as CD45R/B220 or CD11b identifies Flk-2-positive subpopulations of B cells or myeloid cells.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse bone marrow cells at ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.
• Fluorochrome: PerCP-eFluor® 710 (Emission 710 nm, Excitation 488 nm, Blue laser). Recommended 710/50 bandpass filter (695/40 acceptable).
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using:
  • IC Fixation Buffer (Product #00-8222-49)
  • 1-step Fix/Lyse Solution (Product #00-5333-54)
    Clone-specific performance should be empirically verified.
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FLT3 is a tyrosine protein kinase receptor that binds fms-related tyrosine kinase 3 ligand, inducing receptor dimerization and autophosphorylation. This activation triggers downstream signaling pathways regulating apoptosis, proliferation, and differentiation of hematopoietic cells. Constitutive activation mutations are associated with acute myeloid and lymphoblastic leukemia.
