Product Specific Information
This antibody reacts with an extracellular epitope of CD22 (BL-CAM), a 130 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed in the cytoplasm of pro-B and pre-B lymphocytes, and on the surface of mature and activated B lymphocytes. It is lost on plasma cells, peripheral blood T lymphocytes, granulocytes, and monocytes.
Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in both the cytoplasm and membrane of B-lymphocytes. CD22 appears early in B-cell differentiation, similar to CD19. Unlike other B-cell markers, CD22 membrane expression is limited to mature B cells (CD22⁺) and lost in plasma cells (CD22⁻), making it useful in phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and is also positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 binds preferentially to α2,6-linked sialic acid; this recognition site can be masked by cis interactions on the same cell surface. Ligand-induced tyrosine phosphorylation during immune response suggests a regulatory role in B-cell antigen receptor signaling. CD22 may act as both a positive regulator via interaction with Src family tyrosine kinases and as an inhibitory receptor by recruiting cytoplasmic phosphatases that block signal transduction through dephosphorylation.
