Product Specific Information
This antibody reacts with an extracellular epitope of CD22 (BL-CAM), a 130 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed in the cytoplasm of pro-B and pre-B lymphocytes, and on the surface of mature and activated B lymphocytes. It is lost on plasma cells, peripheral blood T lymphocytes, granulocytes, and monocytes.
Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in both the cytoplasm and cell membrane of B-lymphocytes. CD22 appears early in B-cell differentiation, similar to CD19, and is expressed on mature B cells (CD22⁺) but not plasma cells (CD22⁻). It is useful in phenotyping mature leukemias and is strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid. Its sialic acid recognition site can be masked by cis interactions with sialic acids on the same cell surface. Upon ligand-induced tyrosine phosphorylation, CD22 is involved in regulation of B-cell receptor signaling. It can act as both a positive regulator via Src family tyrosine kinases and as an inhibitory receptor by recruiting cytoplasmic phosphatases that block signal transduction.
