This antibody reacts with an extracellular epitope of CD22 (BL-CAM), a 130 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed in the cytoplasm of pro-B and pre-B lymphocytes, and on the surface of mature and activated B lymphocytes. It is lost on plasma cells, peripheral blood T lymphocytes, granulocytes, and monocytes.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein with a molecular weight of 130–140 kDa. It is expressed in both the cytoplasm and cell membrane of B-lymphocytes. CD22 appears early in B-cell differentiation, similar to CD19. Membrane expression is limited to mature B cells (CD22+) and absent in plasma cells (CD22−), making it useful for phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma. It binds preferentially to α2,6-linked sialic acid and plays a regulatory role in B-cell antigen receptor signaling via tyrosine phosphorylation and interactions with Src family kinases and phosphatases.
