Target Information
PAPD5 is a terminal nucleotidyltransferase that catalyzes the transfer of ATP and GTP to RNA 3′ poly(A) tails, creating heterogeneous tails that stabilize mRNAs by preventing deadenylation (PubMed:21788334, PubMed:30026317). It functions as a catalytic subunit of a TRAMP-like complex with poly(A) RNA polymerase activity, involved in post-transcriptional quality control. Polyadenylation with short oligo(A) tails is required for exosome-mediated RNA degradation. PAPD5 operates independently of cofactors for polyadenylation activity (in vitro) (PubMed:21788334, PubMed:21855801). It is essential for cytoplasmic polyadenylation of mRNAs involved in carbohydrate metabolism, including SLC2A1/GLUT1 (PubMed:28383716), and plays roles in histone mRNA degradation, sister chromatid cohesion (PubMed:18172165), and 3′ adenylation of microRNA MIR21 and H/ACA box snoRNAs, influencing their stability and degradation (PubMed:25049417, PubMed:22442037).
