Product Specific Information

  • Immunohistochemistry (PFA fixed):
    Incubate antibody for 30 minutes at room temperature.
    Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in multiple cellular processes including:

  • Ribosome assembly and transport
  • Cytoplasmic/nuclear trafficking
  • Regulation of DNA polymerase α activity
  • Centrosome duplication
  • Regulation of p53

NPM1 continuously shuttles between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. It binds nucleic acids, prevents protein aggregation, protects enzymes from thermal denaturation, and assists in renaturation of denatured proteins.
It is associated with centrosome duplication and serves as a substrate for CDK2/cyclin E.
NPM1 is implicated in several tumor-associated translocations (e.g., NPM-ALK, NPM-RAR, NPM-MLF1) and may mediate oligomerization and oncogenic transformation.
Additionally, it sequesters the tumor suppressor RF in the nucleolus, preventing degradation until needed.
Dysfunction of NPM1 is linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis.
