Product Specific Information
- Tested in direct ELISA (Use antibody at 2–4 µg/mL for coating).
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
NPM1 (Nucleophosmin 1, B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, cytoplasmic/nuclear trafficking, regulation of DNA polymerase α activity, centrosome duplication, and regulation of p53.
It continuously shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones and preventing protein aggregation. NPM1 also protects enzymes during thermal denaturation and assists in renaturation of denatured proteins.
NPM1 is associated with centrosome duplication and acts as a substrate of CDK2/cyclin E. It participates in several tumor-associated fusion proteins (NPM-ALK, NPM-RAR, NPM-MLF1) and is implicated in oncogenic processes.
Dysfunction of NPM1 is linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis.
