Product Specific Information
This HLNC4 monoclonal antibody reacts with human poly (ADP-ribose) polymerase (PARP1), a 116 kDa nuclear enzyme involved in DNA repair.
During apoptosis, caspases -3, -6, and -7 cleave PARP1 after Asp214, producing 85 kDa and 25 kDa fragments. HLNC4 specifically recognizes the 85 kDa cleaved fragment and not the full-length protein.
Immunogen: NH2-GVDEVAKKKSKKEKDC-COOH
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated on Jurkat cells using Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Use at 5 µL (0.06 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®.
Excitation: 405 nm; Emission: 445 nm; Laser: Violet.
Ensure instrument compatibility.
Post-manufacturing filtration: 0.2 µm.
Target Information
Poly ADP-Ribose Polymerase (PARP) uses NAD⁺ as a substrate to transfer ADP-ribose to nuclear proteins.
Cleavage of PARP into its stable 85 kDa fragment is an early marker of apoptosis, mediated by caspase CPP32.
Cleavage occurs between Asp216 and Gly217, conserved across species.
