Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
Thyroid Peroxidase (TPO) is a membrane-bound glycoprotein enzyme essential for thyroid gland function. It catalyzes iodination of tyrosine residues in thyroglobulin and formation of phenoxy-ester bonds between iodinated tyrosines, leading to synthesis of thyroid hormones thyroxine (T4) and triiodothyronine (T3).
Mutations in the TPO gene can cause disorders such as congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Multiple transcript variants exist, though some full-length forms remain undetermined.
Diseases associated with TPO include Thyroid Dyshormonogenesis 2A and Congenital Hypothyroidism.
