- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Thyroid Peroxidase (TPO) is a membrane-bound glycoprotein enzyme essential for thyroid gland function.
It catalyzes the iodination of tyrosine residues in thyroglobulin and the formation of phenoxy-ester bonds between iodinated tyrosines, leading to the synthesis of thyroid hormones thyroxine (T4) and triiodothyronine (T3).
Mutations in the TPO gene are associated with disorders such as congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA.
Multiple transcript variants encoding distinct isoforms have been identified, although some full-length variants remain undetermined.
