Immunohistochemistry (PFA fixed):
Incubate antibody for 30 min at room temperature (RT).
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

Thyroid Peroxidase (TPO) is a membrane-bound glycoprotein enzyme essential for thyroid gland function.
It catalyzes iodination of tyrosine residues in thyroglobulin and the formation of phenoxy-ester bonds between iodinated tyrosines, leading to production of thyroid hormones such as thyroxine (T4) and triiodothyronine (T3).
Mutations in TPO can cause disorders including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA.
Several transcript variants encoding distinct isoforms have been identified, though some full-length forms remain undetermined.
Diseases associated with TPO include Thyroid Dyshormonogenesis 2A and Congenital Hypothyroidism.
