This 1A8 monoclonal antibody reacts with human, mouse, and rat Grim-19. Grim-19 (16 kDa) is expressed in many human tissues, especially in the heart and skeletal muscle, with lower levels in liver, kidney, and brain. Initially described as a nuclear protein, Grim-19 is primarily localized to mitochondria as an essential component of complex I of the electron transport chain. Its expression can be induced by interferon-beta and all-trans-retinoic acid in tumor cell lines, leading to cell death. Grim-19 binds the transactivation domain of STAT3 and inhibits its transcriptional activity.

Applications Reported: Immunoblotting, immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections, and immunocytochemistry.
Applications Tested: Tested by immunoblotting of HeLa cell lysate at ≤5 µg/mL. Antibody should be titrated for optimal performance.
Purity: >90% (SDS-PAGE).

Target Information

GRIM-19 (Gene associated with Retinoid-IFN-induced Mortality) is a novel gene identified to regulate cell death. Antisense expression of GRIM-19 confers resistance to IFN/RA-induced death by reducing intracellular GRIM-19 protein levels, while overexpression enhances cell death in response to IFN/RA. GRIM-19 is primarily nuclear, and its expression is induced by IFN/RA combination, indicating it functions as a cell death-regulatory molecule.
