Target Information

The Factor XIII A subunit gene encodes a component of coagulation factor XIII, the final zymogen activated in the blood coagulation cascade. Plasma factor XIII is a heterotetramer of two A subunits (catalytic) and two B subunits (carrier). Platelet factor XIII consists of only two A subunits identical to plasma origin.

Upon thrombin cleavage and in the presence of calcium ions, plasma factor XIII releases its B subunits to form the active enzyme factor XIIIa, which functions as a transglutaminase. This enzyme catalyzes gamma-glutamyl-epsilon-lysine crosslinking between fibrin molecules, stabilizing the fibrin clot, and also crosslinks alpha-2-plasmin inhibitor or fibronectin to fibrin alpha chains.

Deficiency of factor XIII is categorized as:

  • Type I deficiency: Lack of both A and B subunits
  • Type II deficiency: Lack of A subunit only

Such deficiencies can lead to prolonged bleeding, impaired wound healing, and recurrent miscarriage.
