MA3-007 detects several members of the heat shock protein 70 kDa (HSP70) family, including HSP70, HSC70, GRP78, and HSP72, across multiple species such as yeast, rat, Drosophila, fish, mouse, avian, amphibian, and human samples.
This antibody has been successfully used in:

By Western blot, MA3-007 detects proteins ranging from ~70 kDa to ~78 kDa, representing different members of the HSP70 family. Two-dimensional gel electrophoresis is required to distinguish heat-induced forms from constitutively expressed counterparts. Immunofluorescence staining in heat-shocked HeLa cells shows cytoplasmic localization.
Epitope mapping indicates that MA3-007 recognizes an epitope located between amino acids 122–264 of human HSP70, corresponding to the ATP-binding region. This is the first monoclonal antibody known to react with the ATP-binding region and the amino terminus of HSP70.
Target Information
HSP70 is a heat shock protein expressed in response to various biological stresses, including heat. The HSP70 family includes highly conserved proteins induced by stress across all examined organisms.
Human HSP70 family members:
- HSP70: Inducible and constitutively expressed in primate cells
- HSP72: Induced exclusively under stress conditions
- HSC70: Constitutively expressed, involved in clathrin-coated vesicle uncoating
- GRP78 (BiP): Glucose-regulated protein localized in the endoplasmic reticulum
- HSP75 (p75): Found in mitochondria
HSP70 stabilizes proteins against aggregation and assists folding of newly translated proteins. It also participates in the ubiquitin-proteasome pathway through interaction with AU-rich element RNA-binding protein 1. The HSP70 gene resides in the MHC class III region with two closely related genes encoding similar proteins.
