Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse major histocompatibility complex (MHC) class II molecules, recognizing both I-A and I-E subregion-encoded glycoproteins (I-A^b, I-A^d, I-A^q, I-E^d, I-E^k, but not I-A^f, I-A^k, or I-A^s).
It detects a polymorphic determinant present on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice with H-2^b, H-2^d, H-2^q, H-2^p, H-2^r, and H-2^u haplotypes, but not H-2^s or H-2^f.
The M5/114 antibody inhibits I-A–restricted T cell responses of H-2^b, H-2^d, H-2^q, and H-2^u haplotypes.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; ≤0.06 µg per test recommended. A test is defined as the amount of antibody used in a 100 µL staining volume. Optimize titration for best performance.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells such as macrophages, dendritic cells, and B cells.
They bind endocytosed exogenous antigens and present them to T cells. The antigen-binding groove between the MHC class II alpha and beta chains is open at both ends and accommodates peptides 15–24 amino acids long.
HLA and MHC antibodies are crucial in immunopeptidomics for identifying and characterizing neoantigens via high-performance liquid chromatography coupled with tandem mass spectrometry.
