Usage Recommendations

• Centrifuge the conjugate solution briefly before use and use only the supernatant to minimize aggregates and background staining.
• Optimal antibody dilution should be determined empirically for each application.
• Tested by flow cytometric analysis of human PBMCs at ≤0.25 µg/test (100 µL final volume).
• Recommended cell count: 10⁵–10⁸ cells/test.
• Excitation: 590 nm; Emission: 618 nm; Laser: Yellow Laser.
• 0.2 µm filtered post-manufacturing.

Target Information

TIGIT binds with high affinity to the poliovirus receptor (PVR), leading to increased IL10 secretion and decreased IL12B secretion. It suppresses T cell activation by promoting the generation of mature immunoregulatory dendritic cells.
