Product Specific Information

Description:
The 390 monoclonal antibody reacts with mouse CD31 (platelet-endothelial cell adhesion molecule-1, PECAM-1, gpIIa). This 130–140 kDa surface protein is expressed by endothelial cells and at low levels on all leukocytes and platelets. CD31 mediates homotypic cell adhesion and participates in signal transduction and cell–matrix interactions.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow cells. Recommended use ≤0.06 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal dilution should be titrated empirically.

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is excited by a 405 nm violet laser. Ensure instrument compatibility before use.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54).
Note: BV711 is not compatible with methanol-based fixation.

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling through ITIM motifs. It is expressed throughout the vascular system, primarily at endothelial junctions. The N-terminal Ig-like domain mediates homophilic binding, critical for cell-cell interactions. CD31 functions in integrin-mediated adhesion, transendothelial migration, angiogenesis, apoptosis, immune regulation, and thrombosis.
