Product Specific Information

The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed on early T/B progenitors and activated mature lymphocytes. It associates with CD122 and CD132 to form the high-affinity IL-2 receptor, playing a key role in lymphocyte differentiation and proliferation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
Use 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is optimized for cytometers with a 405 nm violet laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dyes (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

• Super Bright Complete Staining Buffer (#SB-4401-42) or
• Brilliant Stain Buffer™ (#00-4409-75)
  to minimize non-specific interactions.

Light Sensitivity:
BV711 is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored in:

• IC Fixation Buffer (#00-8222-49) or
• 1-step Fix/Lyse Solution (#00-5333-54)
  for up to 3 days at 4°C (dark).
  Not compatible with methanol-based fixation.

Excitation/Emission: 407 nm / 713 nm (Violet Laser)

Target Information

CD25 (IL2RA) encodes the interleukin-2 receptor alpha chain, a cytokine receptor involved in T and B lymphocyte proliferation. Together with IL2RB and IL2RG, it forms the high-affinity IL-2 receptor complex. IL2RA expression regulates immune responses and lymphocyte activation. Mutations can cause IL-2 receptor alpha deficiency, associated with immune dysregulation.
