Product Specific Information

The M1/69 monoclonal antibody reacts with the mouse CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa molecule is GPI-anchored and expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. Expression varies due to glycosylation differences across cell types and is useful for identifying B-cell developmental stages in mouse bone marrow. CD24 also binds to P-selectin (CD62P).

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Optimize titration for best results.

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is suitable for instruments with a 405 nm violet laser. Protect from light as the dye is photo-sensitive.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Fixation:
Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54).
BV711 is not compatible with methanol-based fixation.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B cells in bone marrow to mature surface Ig-positive B cells. Expression is low or absent in plasma cells. CD24 is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity.
