Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R), a type I glycoprotein (~75–80 kDa). It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

Biological Role:
IL-7 binding triggers signal transduction via tyrosine kinase pathways including Jak/STAT. IL-7 is essential for lymphocyte development and T-cell homeostasis. CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs), making CD127 a valuable marker for identifying functional Tregs.

Blocking Information:
Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Applications Reported:
Validated for flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume).

Fluorochrome Information:
Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is designed for cytometers with a 355 nm UV laser.
Use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-Step Fix/Lyse Solution (Product #00-5333-54).

Target Information

CD127 (Interleukin-7 Receptor alpha)
A glycoprotein involved in lymphopoiesis regulation. The IL-7 receptor is a heterodimer of alpha and gamma chains.

• Function: Promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature T cells.
• Signaling: The gamma(c) chain activates signal transduction; the alpha chain mediates specific signaling events.
• Clinical relevance: Mutations or dysfunctions are associated with severe combined immunodeficiency (SCID).
