Description:
This YKIX716.13 monoclonal antibody reacts with canine CD45 (leukocyte common antigen). CD45 is expressed by all hematopoietic cells except red blood cells and platelets and exists as multiple isoforms due to alternative splicing. The C-terminal domain exhibits tyrosine phosphatase activity that enables signaling by T and B cell receptors following activation.

This antibody recognizes all isoforms of CD45. When used with YKIX753.22, it has been reported to promote cell depletion via antibody-dependent cell-mediated cytotoxicity.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal canine peripheral blood cells. Use at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in 100 µL final volume. Cell number may vary from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, emits at 445 nm, and is excited with the 405 nm Violet laser. Ensure the instrument can detect this fluorochrome.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It is absent on non-hematopoietic cells and tissues. CD45 is essential in lymphocyte development and antigen signaling, regulating Src-family kinases and cytokine receptor signaling. It also modulates integrin-mediated signal transduction, apoptosis, and various immune functions. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
