Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse major histocompatibility complex (MHC) class II glycoproteins encoded by I-A and I-E subregions (I-A^b, I-A^d, I-A^q, I-E^d, I-E^k).
It detects polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice with H-2^b, H-2^d, H-2^q, H-2^p, H-2^r, and H-2^u haplotypes, but not H-2^s or H-2^f.

This antibody inhibits I-A-restricted T cell responses of H-2^b, H-2^d, H-2^q, H-2^u haplotypes.
It is validated for flow cytometric analysis of mouse splenocytes and can be used at ≤0.125 µg per test (100 µL final volume).
Cell number should be empirically determined (10⁵–10⁸ cells/test).
Careful titration is recommended for optimal assay performance.

eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm (Violet laser).
Ensure instrument compatibility for detection.
Post-manufacturing filtration: 0.2 µm.


Target Information

MHC class II molecules are transmembrane glycoproteins expressed on antigen-presenting cells (macrophages, dendritic cells, B cells).
They bind endocytosed exogenous antigens and present them to T cells.
The antigen-binding groove between MHC class II α and β chains is open at both ends and accommodates 15–24 amino acid residues.
HLA and MHC antibodies are essential in immunopeptidomics for neoantigen identification and characterization via LC-MS/MS.
