Product Specific Information
1 µg/mL of MA5-45459 was sufficient for detection of Shank3 in 10 µg COS cell lysate transiently transfected with Shank3 by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.
Detects approximately 190 kDa. No cross-reactivity against Shank1 or Shank2.
Target Information
SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance. It connects receptors of the postsynaptic membrane, including NMDA-type and metabotropic glutamate receptors, through complexes with GKAP/PSD-95 and HOMER, and links to the actin-based cytoskeleton.
SHANK3 regulates actin dynamics, neuronal growth cone motility, and NMDA receptor signaling, and modulates GRIA1 and GRM5/MGLUR5 expression to control synaptic transmission and plasticity. It may be required at an early stage of synapse formation and can be inhibited by IGF1 to promote synapse maturation. (Uniprot)
