1 µg/mL of MA5-45460 was sufficient for detection of Shank3 in 10 µg COS cell lysate transiently transfected with Shank3 by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody. Detects approximately 190 kDa. No cross-reactivity against Shank1 or Shank2.
Target Information
SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance. It links receptors of the postsynaptic membrane, including NMDA-type and metabotropic glutamate receptors, via complexes with GKAP/PSD-95 and HOMER, respectively, and the actin-based cytoskeleton. SHANK3 regulates actin dynamics through interactions with Arp2/3 and WAVE1 complexes, influencing neuron growth cone motility and NMDA receptor signaling. It also modulates GRIA1 exocytosis and GRM5/MGLUR5 signaling, contributing to synaptic transmission and plasticity. SHANK3 may play a role in early synapse formation and is inhibited by IGF1 to promote synapse maturation. (Uniprot)
